应用正交试验建立树莓组织培养快繁体系

- 为促进树莓工厂化育苗,本试验采用俄罗斯引进的树莓品种“双季2381”和“单季1132”为材料,以茎段为外植体,应用正交试验对最佳腋芽萌发培养、丛生芽增殖及组培苗生根培养基进行筛选,建立树莓快繁技术体系。结果表明:腋芽萌发最佳培养基“双季2381”为6-BA 1 mg?L-1 + NAA 0.3 mg?L-1,“单季1132”为6-BA 1.0 mg?L-1 + TDZ 0.5 mg?L-1 + NAA 0.5 mg?L-1,其萌芽率分别为91.7%和92.3%,且腋芽萌发快、叶色绿。丛生芽增殖最佳培养基:“双季2381”为6-BA 2.0 mg?L-1 + TDZ 0.5 mg?L-1 + NAA 0.1 mg?L-1,“单季1132”为6-BA 1.0 mg?L-1 + TDZ 1.0 mg?L-1 + NAA 0.3 mg?L-1 ,其增殖系数分别为6.7和6.2,且丛生芽增殖快,叶色绿。组培苗生根最佳培养基“双季2381”和“单季1132”均为1/2MS + IBA 0.2 mg?L-1,其生根率分别达96.9%和95.1%,且自培苗生根快,叶色绿。
