- 为加速翠菊新品种的培育和扩繁,研究利用种子消毒接种获得的无菌苗作为外植体,以MS、1/2MS为基本培养基,添加不同浓度的6-BA、NAA依次进行愈伤组织诱导、愈伤组织分化、不定芽生根。结果表明:茎段为再生体系的最佳外植体,愈伤诱导培养基为MS+2.0 mg?L-1 6-BA+0.2 mg?L-1 NAA,诱导愈伤组织分化不定芽的最佳培养基为 MS+1.0 mg?L-1 6-BA+0.1 mg?L-1 NAA,不定芽最佳生根培养基为1/2MS。叶片、叶柄最佳愈伤诱导培养基MS+4.0 mg?L-1 6-BA+0.2 mg?L-1 NAA,愈伤分化培养基只分化不定根。
- Abstract:
- In order to accelerate the cultivation and propagation of Callistephus chinensis,taking aseptic seedlings obtained by seed disinfection as explants,MS and 1/2MS as the basic medium,different concentrations of 6-BA,NAA were used to induce the formation and differentiation of callus,following the adventitious buds grew roots.The results showed that the stem segment was the optimal explant for regeneration system,the optimal callus induction medium was MS+2.0 mg.L-1 6-BA+0.2 mg.L-1 NAA,the optimal medium was MS+1.0 mg.L-1 6-BA+0.1 mg.L-1 NAA to induce the proliferation of adventitious buds from callus,the best rooting medium of adventitious buds was 1/2MS.The best callus induction medium for leaves and petioles was MS+4.0 mg.L-1 6-BA+0.2 mg.L-1 NAA,callus differentiation medium was only for forth adventitious roots.
