- 对美国红枫组织培养中外植体灭菌、褐化预防及外植体启动进行了试验研究。结果表明:以美国红枫半木质化茎段为外植体,用1%次氯酸钠灭菌4 min,外植体的成活率最高为82.22%;在外植体培养初期每隔2 d转移至新鲜培养基中,2~3次后可以彻底去除褐化现象;最佳的启动培养基为MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1,外植体抽芽率可达92.2%。
- Abstract:
- The America Red Maple(Acer rubrum)was taken as material cultured in vitro to study sterilization techniques,browning prevention and explants initial culture.The results showed that using haif-lignification stems as explants,the highest survival rate of the explants was 82.22% after the explant were sterilized in 1% NaClO for 4 min.Transfer explants into fresh medium every 2 d at primary stage,browning can be eliminated then after 2~3 times.The MS medium containing 1.0 m·L-1 6-BA and 0.1 m·L-1 NAA was most efficient in the initial culture of Acer palmatum atropurpureum,sprouting rate of explants was 92.2%.
