- 采用BSA 法对垦农18(高油, 23. 21 %)与黑农小粒豆(低油, 19. 0 %)的杂交F2 :3 群体进行RAPD 分析。用200 个随机引物对构建的高油DNA 池和低油DNA 池进行扩增, 有46 个引物产生了RAPD 扩增产物, 共获得315 个片断, 其中产生多态性片段的随机引物为4 个, 同高油含量相关的特征性条带分别为S56-900bp 、S61-600bp 、S107-600 /450 /370bp 、S134-450 /500bp 。这些标记平均可以区分F2 :3群体中87. 1 %的高油与低油单株, 重复性好, 可以用作高油含量的分子标记。
- Abstract:
- With Bulked Seg reg ation Analy sis , RAPD metho d w as employed to analy ze genome DNA of F2 :3 populat ion obtained by cro ssing Kennong 18(oil co ntent , 23. 21 %)w i th Heinongx iaolidou(oil content , 19. 0 %). Tw o hundred random primers w ere used to amplify betw een thehig h and low oil co ntent DNA poo ls , amo ng w hich there w ere 315 amplified f ragment s by 46primer s that produced RAPD product and w e found there w ere 4 random prime rs produced polymorphicbands. 4 markers rela ted w ith oil co ntent w ere S56-900bp 、S61-600bp 、S107-600 /450 /370bp 、S134-450 /500bp respectively. T hese markers could separate 87. 1 % high or low plant s inF2 :3 po pulatio n. The result s showed very stable and repeatable. The RAPD markers was useful to assistantthe selection fo r high oil content soybean plant s in soybean hybrid breeding.
