多脂鳞伞的抗氧化活性及对小鼠免疫功能的影响

- 多脂鳞伞是一种具有经济价值的食药两用真菌,为了探讨多脂鳞伞抗氧化活性及对小鼠的免疫功能,使用石油醚、二氯甲烷、乙酸乙酯与甲醇对多脂鳞伞干燥子实体进行提取得到相应组分,并进行清除ABTS·、DPPH·自由基的研究;用煎煮法获得多脂鳞伞水提液,对42~56日龄昆明小鼠进行为期20 d灌胃给药,测定其血清IL-2、SOD、IFN-γ及GSH-Px等生化指标。结果表明,在抗ABTS·、DPPH·自由基试验中,多脂鳞伞乙酸乙酯提取组分对DPPH·自由基清除率达到91.45%,在质量浓度为1.0 mg·mL-1时,ABTS·自由基清除率为65.24%,表现出良好的抗氧化活性;多脂鳞伞水提液对小鼠血清中IL-2、SOD、IFN-γ及GSH-Px的含量均有显著增加,并且呈现浓度依赖性,表现出良好的调节免疫作用,说明多脂鳞伞具有良好的抗氧化及提高机体免疫力的作用。
- Abstract:
- The fruiting bodies of Pholiota adiposa were cultivated as materials, and the corresponding extracts were obtained with petroleum ether, methylene chloride, ethyl acetate and methanol as solvents. The antioxidant activities of DPPH· and ABTS· free radical scavenging rate were studied. The results showed that, the ethyl acetate extract of polylipidium had DPPH· radical scavenging ability, and its clearance rate was 91.45%. The ethyl acetate extract had the strongest scavenging ability for ABTS· radical, and the scavenging rate of ABTS· radical was 65.24% when the mass concentration was 1.0 mg·mL-1. Kunming mice were given different doses of water extract for 20 days. The levels of cytokines such as IL-2, SOD, IFN-γ and GSH-Px in serum were determined to evaluate the immune function of Kunming mice. The results showed that the contents of IL-2, SOD, IFN-γ and GSH-Px in the serum of the mice treated with different concentrations of polyliposis were significantly increased, and all of them had good immunomodulatory and anti-oxidative stress effects.
