西洋参内生真菌分离鉴定及生防特性

- 为促进西洋参病害的生物防治研究,通过采用纯培养方法分离纯化西洋参内生真菌,获得58株内生真菌,通过形态学鉴定和显微观察初步确立了16株菌,提取纯化出其中13株真菌DNA,选用真菌通用引物ITS1和ITS4对DNA进行ITS特异性扩增,提交测序分析,构建系统发育树。确定其分属于:镰孢属(Fusarium)、毛束霉属(Trichurus)、葡萄孢属(Cinerea)、亚黑团孢属(Periconiella)、棘壳孢属(Pyrenochaeta)、链格孢属(Ilternarsa)。同时对菌株的生防作用进行研究,其中14株针对立枯病、根腐、茎腐、液斑水浸木4种病原菌表现出不同程度的拮抗性,占分离菌株的87.5%。试验表明西洋参内生真菌多样性丰富。
- Abstract:
- In order to promote the biological control research on the disease of Panax quinuefolium,58 strains were isolated form Panax quinquefolium by using the methods of inserting tissue and spreading milling liquid.According to their colony characters and microscopic morphological characters,16 strains are identified.We extract the DNA of 13 strains and amplify the ITS sequence by using the PCR apparatus.We contrast the ITS sequence and establish phylogenetic tree.There are belong to 6 genus:Fusarium,Trichurus,Cinerea,Pyrenochaeta,Ilternaria,Periconiella.14 strains showed different degrees of resistance for Rhizoctonia solani Kuhn,ceitocybe bescens,stem rot,leaf spot,accounting for 87.5% of the isolates.Panax quinquefolium endophytic fungi have a rich diversity.
