- 为提高红阳猕猴桃组织培养效率,以红阳猕猴桃带芽嫩茎段和再生苗叶片为外植体,通过组培快繁试验,对影响红阳猕猴桃嫩茎段腋芽萌发和再生苗不定芽再生的生长调节物质的用量和作用进行分析筛选。结果表明:红阳猕猴桃最适宜诱导腋芽萌发的培养基为WPM+1 mg?L-1 6-BA+0.1 mg?L-1NAA和WPM+ 1 mg?L-16-BA+0.2 mg?L-1NAA,在此条件下带腋芽茎段萌芽率达100%;再生苗叶片不定芽诱导的最佳培养基为WPM+1 mg?L-1 6-BA+0.2 mg?L-1NAA+0.2 mg?L-1 2,4-D;最适合红阳猕猴桃不定芽增殖的培养基为WPM+0.2 mg?L-1NAA+0.2 mg?L-1 2,4-D。
- Abstract:
- In order to improve the tissue culture efficiency of Actinidia chinensis var.rufopulpa Liang et Ferguson,the stems and leaves were used as explants to investigate the tissue culture and propagation technique.Effects of growth regulating substance on bud sprouting and adventitious buds regeneration were also tested.The results indicated that the most suitable medium for the induction of axillary buds was WPM+1 mg?L-1 6-BA+0.1 mg?L-1NAA and WPM+1 mg?L-16-BA +0.2 mg?L-1NAA,and the bud rate of the axillary buds was 100% under this condition.The optimal medium for the induction of the leaf of regeneration seedlings was WPM+1 mg?L-1 6-BA +0.2 mg?L-1NAA+0.2 mg?L-1 2,4-D; The most suitable medium for adventitious bud proliferation was WPM+0.2 mg?L-1NAA+0.2 mg?L-1 2,4-D.
