- 采用组织块贴壁培养法分离获得野猪输卵管上皮细胞,并进行传代、冻存和复苏,并对其生物学特性进行观察研究。结果表明:组织块在培养2 d开始有细胞爬出,7 d左右可长满全皿,原代细胞形态呈菱形,其中混有少量成纤维细胞。传代2~3次,细胞生长状态最好,4代后细胞退化;细胞经冷冻保存再解冻后细胞死亡率上升。通过此方法能够获得较纯的野猪输卵管上皮细胞,为野猪资源保存和进一步的研究提供技术支持。
- Abstract:
- The method of tissue blocks adherent culturewas used to isolate wild boar oviduct epithelial cells,and the cells were passed and cryopreservation and anabiosis,their biological characteristics were observed.The results showed that:the cells grew out from tissue blocks on the next day,and covered dish about 7 days.Primary cells were diamond and there were a few of fibroblast cells.The cells had fine growth state when they were passed 2 to 3 times,and degenerated after 4 times passed.The mortality rate increased when cells were cryopreserved and then thawed.The results showed that pure wild boar oviduct epithelial cells could be obtained by this method,and the method could provide technical support for the resource preservation and further study of wild boar.
