- 利用同源克隆及RACE的方法,从白背飞虱Sogatella furcifera中克隆获得热激蛋白HSP90基因的全长cDNA序列(Genbank登录号:JQ743628),该基因全长为2 707 bp,包含421 bp的3′非编码区域(UTR)和93 bp的5′UTR,开放阅读框(ORF)为2 193 bp,编码730个氨基酸。该基因预测的蛋白分子量为83.85 kD,等电点为4.94,无信号肽、糖基化位点及跨膜结构。在N端具有HSP90基因保守的ATPase结构,含有HSP90家族的C末端的保守序列MEEVD。从进化分析结果看出,白背飞虱的HSP90与其它昆虫的HSP90蛋白的同源性较高,与褐飞虱相比较,同源性高达98%。HSP90基因的克隆和比较分析为进一步深入研究白背飞虱的抗逆机理、耐受性及进化具有重要意义。
- Abstract:
- The heat shock protein 90(HSP90)gene from Sogatella furcifera was cloned by homology cloning strategy and technique of Rapid Amplification of cDNA Ends(RACE).The full-length cDNA of S.furcifera HSP90(Sf HSP90) is 2 707 bp(Genbank number:JQ743628)and has an open reading frame of 2 193 bp,a 3 UTR(untranslated region)of 421 bp and a 5UTR of 93 bp.The estimated molecular weight of HSP90 is 83.85 kD and pI is 4.94.There are no glycosylation sites,putative transmembrane domain and signature peptide.It contains ATPase conservative domain in N-site and MEEVD signature sequence on C-site.Compared with Nilaparvata lugens,they are 98% identities and Phylogenetic analysis shows that these two species are most closely related.Cloning and comparison analysis of HSP90 genes from S.furcifera could be useful in the studies of anti-stress mechanism,tolerance improvement breeding and evolution for these animals.
