- 连香树是连香树科连香树属、第三纪古热带的孑遗植物,为我国珍稀濒危二级保护树种,具有极高的科研、经济及观赏价值。试验对连香树无菌体系的建立及植株再生技术进行了研究。结果表明:(1)连香树外植体的最佳消毒方法为0.1%升汞消毒12 min;(2)芽增殖最佳培养基:MS+6-BA 1.0 mg·L-1+NAA 0.5 mg·L-1+GA3 2.0 mg·L-1+蔗糖30 g·L-1+琼脂7.0 g·L-1,pH 6.0;(3)愈伤诱导的最佳培养基:MS+6-BA 0.1 mg·L-1+2,4-D 2.0 mg·L-1+蔗糖30 g·L-1+琼脂7.0 g·L-1,pH 6.0;(4)生根培养基:1/2MS+IBA 1.0 mg·L-1+6-BA 0.1 mg·L-1+蔗糖20 g·L-1+琼脂6.8 g·L-1,pH 6.0。
- Abstract:
- Cercidiphyllum japonicum is a relict species of tertiary ancient tropical plant,which has been listed the national second-grade protected kind and possesses researched economic and appreciatory value.In this paper the plant regeneration of Cercidiphyllum japonicum was researched.The result showed that the best sterilization methods was using 0.1% HgCl2 for 12 min;MS+6-BA 1.0 mg·L-1+NAA 0.5 mg·L-1+GA32.0 mg·L-1+30 g·L-1 sucrose+7.0 g·L-1 agar,pH 6.0 was optimum for reduplication;MS+6-BA 0.1 mg·L-1+2,4-D 2.0 mg·L-1+30 g·L-1 sucrose+7.0 g·L-1 agar,pH 6.0 was optimum to induce callus;1/2MS+IBA 1.0 mg·L-1+6-BA 0.1 mg·L-1+20 g·L-1 sucrose+6.8 g·L-1 agar,pH 6.0 was optimum medium for growing root.
