- 以西方蜜蜂幼虫为材料,采用玻璃化冷冻方法,研究了蜜蜂幼虫冷冻技术。结果表明:从多种抗冷冻保护液中筛选出的适宜蜜蜂幼虫的冷冻保护液为15%甘油+0.2 mol·L-1蔗糖+0.2 mol·L-1木糖+3%聚乙二醇+15%乙二醇。冷冻剂类为纱布包裹型。冷冻程序:预冷时间为5 min,距液氮高度2~7 cm,降速率为1 cm·min-1。解冻液为生理盐水+葡萄糖。此方法冷冻幼虫解冻后成活率为10%左右。
- Abstract:
- Bee larva freezing technology is based on Apis mellifera which makes reference to the embryo freezing technology of large livestock,using vitrification freezing technology.After repeated experiments,the suitable bee larva freezing protective liquid which was selected from a lot of anti-freeze protective liquid was:15% glycerol+0.2 mol·L-1sucrose+0.2 mol·L-1 xylose+3%polyethyleneglycol+15%glycol.The type of cryogen was gauze wrapping.Frozen procedure:pre-cooling time was 5 min,the height above the liquid nitrogen surface was 2~7 cm,the decreasing rate was 1cm·min-1.The thawing solution was physiological saline+ glucose.The survival rate of the freezing larva was about 10% by using this method.
