- 以美洲南蛇藤幼嫩的顶芽或带腋芽的茎段为外植体, 进行外植体消毒、增殖培养、瓶内生根和过渡移栽研究, 结果表明:外植体用75%的乙醇消毒20 s , 0 .1 %的HgCl2 消毒2.0 min 效果最佳;适宜的增殖培养基为M S+6-BA 1 .0mg ·L-1(单位下同)+IBA 0 .2 +3.0 %蔗糖;壮苗培养基为MS +6-BA 0.1 + IBA 0 .05 +3%蔗糖;适宜的生根培养基为1/ 2MS +IBA 0.5+1 .5 %蔗糖, 生根率82 .0 %;最佳过渡移栽基质为蛭石∶珍珠岩=2∶1, 成活率为98 .3 %。
- Abstract:
- Taking top buds and stems with axillary buds as the e xplants , the study of disinfection , pro life ratio n culture, r oo ting in vitro and t ransition cultiva ting were conducted.The results w ere as fo llow s:the most suitable disinfecttime pr operly wa s 20 s by alcohol and 2 min by HgCl2 respectively ;pro liferatio n culture media wa s MS +6-BA1.0mg ·L-1 +IBA0.2 +Sucr ose 3 %;ha rde ning-off culture media wa s MS +6-BA0.1 + IBA0.05 +Sucro se3%;r oo ting cultur e media w as 1/ 2MS +IBA 0 .5 + Sucro se 1.5% and its r oo ting ratio w as 82.0%;transitio n cultivating media was pearlite and peat , its rate wa s 2 ∶1, sur viv e ratio wa s 98.3%.
