丁酸钠对玉米种子萌发和α-淀粉酶活性及其基因表达的影响

- 为深入研究玉米种子萌发的表观调控机制,用梯度浓度(0,0.5,1.0,5.0和10.0 mmol?L-1)丁酸钠(NaB)溶液处理玉米种子(24、48、72和96 h),比较种子萌发率、根长、根干重、根鲜重,探讨丁酸钠对玉米种子萌发的影响。结果表明:低浓度丁酸钠(0.5 mmol?L-1)处理玉米种子,上述测定的指标均上升;而较高浓度(5和10 mmol?L-1)处理,各指标均有明显的下降。说明不同浓度的丁酸钠对玉米种子萌发具有不同的影响,低浓度促进玉米种子萌发,较高浓度则抑制玉米种子的萌发,且抑制作用具有剂量依赖性。进一步选择0.5和10 mmol?L-1两个浓度的丁酸钠处理玉米种子,测定α-淀粉酶活性和基因转录水平。0.5 mmol?L-1丁酸钠增强α-淀粉酶活性、上调α-淀粉酶基因转录;10 mmol?L-1丁酸钠降低α-淀粉酶活性并下调α-淀粉酶基因转录,推测低浓度丁酸钠上调α-淀粉酶基因的表达可能是其促进玉米种子萌发的机制或部分机制。
