- 为进一步优化百合离体培养条件,以亚洲百合鳞片为外植体,研究不同鳞片部位、不同激素配比对亚洲百合试管鳞茎诱导的影响。结果表明: 鳞茎内层下部分化率最高,鳞茎中层下部鳞片污染率低,是亚洲百合适宜选取的外植体材料。6-BA 0.5 mg?L-1?+NAA 0.1 mg?L-1鳞片分化率最高为75.00%,鳞片分化数最高的激素配比为6-BA 2.0 mg?L-1?+NAA 0.1 mg?L-1,分化数为3.05。在6-BA和NAA配合使用的情况下,6-BA 2.0 mg?L-1+NAA 0.1 mg?L-1增殖效果最好,增殖数为13.4个。百合试管鳞茎膨大的理想激素浓度配比为MS+6-BA 2.0 mg?L-1?+NAA 0.1 mg?L-1+ PP3334.0 mg?L-1。
- Abstract:
- The Asiatic hybrid lily (Lilium?spp.) was used as explant to study the effects of different scales and different hormone combinations on the induction of bulblets.The results showed that the differentiation rate of the base part of the inner bulb was the highest,and that the base part of the middle bulb has the lowest pollution rate,which was suitable explant for tissue culture of Asiatic hybrid lily.The best differentiation rate was 75.00% on MS medium supplemented with combinations of 6-BA 0.5 mg?L-1?and NAA 0.1 mg?L-1.And the best differentiation number was 3.05 on MS medium supplemented with combinations of 6-BA 2.0 mg?L-1? and NAA 0.1 mg?L-1.The proliferation rate of bulblet was up to 13.4 on MS medium supplemented with combinations of 6-BA 2.0 mg?L-1? and NAA 0.1 mg?L-1.The best hormone concentration ratio of lily bulb enlargement was MS+6-BA 2.0 mg?L-1? +NAA 0.1 mg?L-1+PP333?4.0 mg?L-1.
