- 为获得大穗结缕草离体繁殖的最佳条件,以大穗结缕草不同器官作为外植体,MS为基本培养基,研究了不同消毒方法对外植体除菌效果的影响,不同外植体以及不同激素组合对愈伤组织诱导及分化的影响。结果表明:在光强1 500~2 000 lx,16 h光照/8 h黑暗,(23±2)℃培养条件下,大穗结缕草外植体,经70%酒精浸泡45 s,0.1%HgCl-2浸泡8 min,污染率低,易于愈伤组织的诱导。以种子胚作为外植体最有利于愈伤组织的培养,愈伤组织诱导的最佳培养基是MS+2,4-D(2.0 mg?L-1)+6-BA(0.2 mg?L-1)+TDZ(1.0 mg?L-1)。
- Abstract:
- In order to identify the best conditions on rapid propagation of Zoysia macrostachya,together with MS as basic medium,different methods of disinfection of explants were studied,the effect of the different types explants (seeds,leaves of Z.macrostachya as explants) and different hormone combinations on induction and differentiation of callus was researched.The results showed that in the condition of light intensity was 1 500~2 000 lx for 16 h light / 8 h dark,(23±2)℃,disinfected with being immersed in 70% alcohol for 45 s,being washed with aceptic water 3 times,being immersed in 0.1% HgCl-2 for 8 min,being washed with aceptic water 3 times,seed embryo are easy to callus growth with low contamination rate. Using seed embryo as explant,the best induction medium were MS+2,4-D(2.0 mg?L-1)+6-BA(0.2 mg?L-1) +TDZ(1.0 mg?L-1).
