- 以水稻粳稻品种空育131、垦稻12 为材料, 用农杆菌共转化的方法将质粒p1300 和p13W8 导入受体材料。结果显示:两种混合共转化菌液按体积比1∶1 时共转化效率最高, 植株转化效率达4.08 %和2 .63%。将转化植株进行PC R 鉴定, 13 株转基因植株均能检测到质粒p1300 的信号, 4 株检测到质粒p13W8 的信号, 反义蜡质基因的阳性株占转基因植株总数的30.77 %。
- Abstract:
- Both of the plasmid p1300 and plasmid p13W8 wer e intro duced into J aponica Rice Kongy u 131 byAgr obacte rium-mediated co-transfo rmation , the same tw o plasmid wer e introduced into Kendao 12 by the sameway .The results show ed tha t the co-transfo rmation efficiency w as highest when tw o kinds of Ag robacterium w as1∶1 vo lume , and the co-t ransfo rmatio n efficiency we re 4 .08% and 2 .63%, re spectively .The t ransfo rmed plantswe re sc reened by PCR method , specific fragments o f plasmid p1300 could be screened in 13 transg enic lines , specificfragments o f plasmid p13W8 co uld be screened in 4 transgenic lines .Po sitive Antisense-Wa xy Gene plantswe re 30 .77 % of all transfo rmed plants.
