羊草幼穗组织培养与再生体系建立的研究

- 为了优化羊草组织培养与再生体系,本试验以6个羊草[Leymus chinensis(Trin.)Tzvel.]新品系的幼穗为外植体,使用添加不同植物激素的MS培养基对羊草幼穗进行愈伤组织诱导、继代、分化等培养,建立高效再生体系。结果表明:诱导培养基中添加2,4-D激素浓度为5.0 mg?L-1时,6个羊草新品系幼穗的诱导率均较高,其中Lc-6出愈率最高,为87.5%,其次是Lc-4,诱导率为81.5%;6个品系愈伤组织通过固-液-固(MS2-MSL2-MS2)交替继代培养,愈伤组织结构与状态得到明显改善,愈伤由疏松变得致密,褐化现象明显减少,愈伤变为乳白色颗粒状,Lc-6和Lc-4的愈伤状态优于其他品系;6个品系愈伤组织在BNKT分化培养基上的分化率显著高于其他2种培养基(P < 0.05),Lc-6分化率最高,为41.25%;6个品系移栽后组培苗成活率均为100%。
