- 为提高甜叶菊的组织培养效率,以甜叶菊幼叶为外植体, 研究75%消毒酒精和0.1%氯化汞的不同消毒时间组合对外植体的消毒效果; 以及不同激素浓度组合对外植体愈伤组织的诱导和芽分化的影响。 结果表明:甜叶菊幼叶外植体材料的最佳灭菌条件为75%消毒酒精30 s,0.1%氯化汞2 min;最佳诱导愈伤组织和分化芽培养基为MS+1.5 mg?L-1 6-BA+0.5 mg?L-1 NAA,此时不定芽增长率为85.7%,外植体适宜的生根培养基为1/2 MS十0.2 mg?L-1IAA,生根率为88.6%。
- Abstract:
- In order to improve the culture efficiency of Stevia rebaudiana Bertoni,taking the young leaves as materials to study the effect different disinfection time of 75% disinfected alcohol and 0.1% mercuric chloride,and different hormone combinations on callus initiation and shooting.The results showed that the optimum sterilization conditions for the leaves of Stevia rebaudiana were 75% sterile alcohol 30 s and 0.1% mercuric chloride for 2 min.The best shooting medium was MS+1.5 mg?L-1 6-BA+0.5 mg?L-1 NAA,and the ratio of inducing was 85.7%.The optimum rooting medium was 1/2 MS+0.2 mg?L-1IAA,and the ratio of rooting was 88.6%.
