- 为了建立绵竹快速繁殖体系,选择绵竹当年生生长健壮的带芽茎段作为外植体,探讨其组织培养中的最佳灭菌方法,并通过正交试验筛选丛芽诱导的最适培养基。结果表明:将外植体加洗洁精清洗后,用自来水冲洗2 h,然后用75%酒精浸泡30 s,0.1%升汞溶液(加入3滴吐温)消毒2次,每次8 min灭菌效果最好,污染率较低,成活率较高。最佳丛芽诱导培养基为:3/4MS+6-BA 3.0 mg?L-1+KT 0.5 mg?L-1 +蔗糖20 g?L-1+琼脂8 g?L-1,丛芽诱导率可达到72.2%。

In order to establish rapid propagation system of Bambusa intermedia,taking the stem with a bud as explants,disinfection method in the tissue culture was investigated,and the optimum medium to induce multiple shoot was optimized by orthogonal experiment.The results showed that the explants cleaned with detergent,washed by tap water for two hours,immersed in 75% alcohol for 30 s and then disinfected by 0.1% mercuric chloride (adding three drops of tween) for eight minute twice could reach good effect of low contamination rate and high survival rate.The optimum? to induce multiple shoot was 3/4MS+6-BA 3 mg?L-1+ KT0.5 mg?L-1 +sucrose 20 g?L-1+ agar 8 g?L-1,induction rate could reach 72.2%.
