- 为亚麻分子标记及分子育种提供可靠的理论依据,利用正交试验设计对亚麻SRAP-PCR反应体系中的4因素(模板DNA、引物浓度、Mg2+浓度、dNTPs浓度)在3个水平上进行正交优化,确立了适合亚麻SRAP-PCR反应的20 μL体系:75 ng模板DNA、0.75 μmol·L-1引物、1.5 mmol·L-1 Mg2+、0.40 mmol·L-1dNTPs和1.5 U Taq DNA聚合酶。
- Abstract:
- In order to provide reliable theory basis for flax molecular marker and molecular breeding,the orthogonal design was used to optimize SRAP-PCR amplification system at 3 levels of 4 factors in flax(DNA template,primer,Mg2+,dNTPs).The most suitable SRAP-PCR reaction system for flax was total 20 μL containing 75 ng DNA template,0.75 μmol·L-1 primers,1.5 mmol·L-1 Mg2+,0.40 mmol·L-1 dNTPs and Taq DNA polymerase 1.5 U.
