- 为解决紫薯栽培中出现退化的问题,以美国紫薯的嫩芽为材料,进行了试管苗培养、试管苗生根继代培养以及对试管苗进行2次15 d 38℃热处理、经热处理后试管苗移栽和定植的研究。结果表明:1/2MS+IAA 0.2 mg·L-1+蔗糖15 g·L-1是美国紫薯嫩芽试管苗培养和试管苗生根继代培养的理想培养基;对试管苗进行2次热处理,部分生长旺盛的试管苗仍能成活并生长出嫩梢,且经热处理生长的嫩梢培养的试管苗容易移栽和定植成活。在相同的栽培条件下经热处理的试管苗比由种薯发出的嫩芽扦插苗的紫薯收获量增产50%左右。
- Abstract:
- In order to solve degradation problems during the cultivation of Ipomoea batatas,tender buds were used as the explant in vitro culture to study rooting,transplanting and field planting of test-tube seedlings.Before transplanting,test-tube seedlings were treated twice under the condition of 38℃ and each treatment lasted 15 days.The results showed that the optimum medium for rooting and subculture was 1/2MS+ IAA0.2 mg·L-1+ sucrose 15 g·L-1.After twice heat treatment,some vigorous growth test-tube seedlings could still survive and grow out of young shoots.The test-tube seedlings which cultivated from the heat-treated young shoots could transplant and field planting easily.Compared with plantlets issued from the shoots cuttings,the harvest yield of heat-treated test-tube seedlings could add fifty percent under the same cultivate condition.
