- 以芫荽基因组DNA为模板,对RAPD扩增反应条件进行优化,以期建立适合芫荽的RAPD的最优反应体系,结果表明:PCR扩增体系(总体积20 μL)为:模板DNA1.0 ng·μL-1,dNTP 0.45 mmol·L-1,随机引物1.3 μmol·L-1,Taq酶0.6 U,Mg2+ 2.0 mmol?L-1,退火温度39℃,循环次数40。
- Abstract:
- Taking the DNA of Coriander sativum as the template,the amplification system was optimized to set up the optimal reaction system.The results showed that the optimum conditions were as follows:20 μL PCR reaction volume,1.0 ng·μL-1 template DNA,0.45 mmol·L-1dNTP,1.3 μmo·L-1 primer,0.6 U Taq DNA polymerase and 2.0 mmol·L-1 Mg2+,39℃annealing temperature and 40 cycles.
