- 通过正交试验对黄粉虫几丁质/壳聚糖的提取工艺进行了优化。结果表明:盐酸浓度、脱乙酰基氢氧化钠浓度和盐酸处理时间对壳聚糖提取有较大影响,其影响程度顺序为盐酸浓度>氢氧化钠浓度>盐酸处理时间。黄粉虫几丁质/壳聚糖的提取最佳工艺条件为10%盐酸室温浸泡2~6 h,7%氢氧化钠100℃煮2.5 h,50%氢氧化钠85℃水浴8 h。在最佳提取工艺下,壳聚糖的提取率达15%,壳聚糖游离胺基大于80%,灰分小于1%,水分4.91%~5.01%。
- Abstract:
- The orthogonalty experiment was used to optimize the extraction conditions of chitin/chitosan from Tenebrio molitor?(L.). The results showed that HCl concentration, NaOH concentration in the process of deacetylation and HCl treating time significantly affect the extraction rate of chitin/chitosan, and the order of effect degree was as follows: HCl concentration>NaOH concentration>HCl treating time. The optimal extraction process were 10% HCl soaking for 2 h to 6 h at room temperature, 7% NaOH boiling for 2.5 h at 100 ℃ and treated with 50% NaOH for 8 h at 85℃. Under these conditions, the extracting rate of chitin/chitosan was 15%. The dissociative amidogen of chitin/chitosan was more than 80%. The ash content of chitin/chitosan less than 1%. The water content of chitin/chitosan was 4.91%~5.01%.
