农杆菌介导北方粳稻转化体系的研究

- 通过农杆菌介导的方法将Bar-Bt 基因(抗除草剂基因和抗虫基因构建在同一载体上的双基因)整合到北方优质粳稻龙稻6 号、松粳9 号等品种中;分别以供试品种的幼胚和成熟胚为外植体诱导愈伤组织, 携带有Bar-Bt 基因双元载体PCAM BIA1301 号的根癌农杆菌EHA105 为载体, GUS 基因的瞬时表达率为衡量指标, 对影响农杆菌介导的龙稻6 号、松粳9 号等品种的转化效率的主要因素进行了研究。结果表明:农杆菌侵染愈伤组织的菌液浓度以D(600nm)= 0 .8 ~ 1.0 较为适宜, 最佳共培养时间为2 ~ 3 d , 预培养时间为5 d , 共培养培养基中添加100 μmo l·L-1 乙酰丁香酮的转化效率有明显提高。

- With juvenile embry o o f g rain-ory za of No rth of China as e xplant , Ag ro bac te rium tumefaciens strainEHA105 car rying Tipla smid PCAMBIA1301 w ith Bar-Bt gene a s vector , and the tr ansient expressio n rate of GUSgene as measuring index , main facto rs that affected tra nsfo rmatio n efficiency of g rain-or yza o f No rth of China mediated by Ag robacterium tumefaciens w ere studied in this a rticle .Results showed tha t the proper concentratio n ofbac teria solution to infect callus w as D(600nm)= 0 .8~ 1 .0 , the most suitable co-cultiva tion time w as 2 ~ 3 days ,pre-cultiv atio n time was 5 day s, and addition o f 100 μmo l·L-1 Aceto sy ring one im pr oved transfo rma tion efficiencyconsider ably.
