银粉背蕨组织培养及无性系建立的研究

- 以银粉背蕨叶柄为材料, 成功地诱导了愈伤组织, 建立起无性系。结果表明:1/2 MS +NH4 H2 PO4 200 mg·L-1 +BA 0.3 mg ·L-1 +IBA 0 .4 mg·L-1 是诱导叶柄形成愈伤组织的理想培养基;1/2 MS 是颗粒状愈伤组织分化培养的理想培养基;1/3 MS + IAA 0 .4 mg ·L-1 是试管苗生根培养的理想培养基;炉灰渣是试管苗移栽的理想基质;移植后的试管苗生长旺盛、根系发达, 秋末收获单株干草重量比野生植株增加14.6%。
- Abstract:
- Callus induction and plant reg ene ratio n from the petio le in Aleuritopteris argentea var .argentea wererepor ted in this re search .The re sults showed that the medium 1/ 2 MS +NH4H 2 PO4 200 mg ·L1 +BA 0.3 mg·L1+IBA 0.4 mg ·L1 wa s the best for inducing callus , medium 1/ 2 MS w as the most suitable fo r bud differe ntiation ,medium 1/3 MS + IAA 0 .4 mg ·L1 w as the optimum for roo ting .The ideal transplantation matrix fo r tube plantletswa s the furnace a shes .The tr ansplanted plantlets in the field g rew vig o rously and had a w ell develo ped r oo tsy stem .The hay w eig ht of the single plant w as 14 .6 % more than that of the wild plant.
