- 以福农39甘蔗为材料，通过预处理、茎尖培养、增殖培养、生根培养、病毒检测、炼苗移栽，以期建立健康种苗繁育体系。结果表明：经50 ℃水浴15 min，在温度为35 ℃、相对湿度为80%、光照强度为1 000 lx的培养箱中培养约7 d，甘蔗茎段侧芽萌发率达97%。在MS+2,4-D1.0 mg?L-1+6-BA1.0 mg?L-1 +AC 5.0 g?L-1培养基中，茎尖均生长良好。在MS+6-BA1.5 mg?L-1+KT0.5 mg?L-1继代培养基中，增殖系数为’13.6。在1/2MS+NAA1.0 mg?L-1生根培养基中，生根率达100%。经检测，组培苗无花叶病和宿根矮化病发生。炼苗移栽后存活率达到86%。由此，建立起福农39甘蔗健康种苗繁育体系。
- Abstract:
- Using Funong 39 sugarcane as material,through pretreating,stem tip culturing,multiplication culturing,rooting culturing,virus detecting,acclimatizing and transplanting,a system of breeding healthy seedling was built.The results showed that staying 15 minutes in the water of 50 ℃,and then staying 7 days in the culturing box that the temperature was 35 ℃,the relative humidity was 80% and the illumination intensity was 1 000 lx,the lateral bud germination rate was 97%.Stem tip grew well in the culture medium of MS+2,4-D 1.0 mg?L-1+6-BA1.0 mg?L-1 +AC5.0 g?L-1.Multiplication rate was 13.6 in the subculture medium of MS+6-BA1.5 mg?L-1+KT0.5 mg?L-1.Rooting rate was up to 100% in the rootig medium of 1/2MS+NAA 1.0 mg?L-1.By detecting,tissue culture seedling was free from mosaic virus and ratoon stunting disease.Survival rate was 86% after acclimatizing and transplanting.A system of breeding healthy seedling for Funong 39 sugarcane was built up.
