空育131和稻花香是黑龙江省很有代表性的寒地栽培品种,也是育种研究的重要材料。为了获取此品种的优质RNA,以经低温避光处理3 d的空育131及稻花香水稻的10日龄水稻苗叶为材料,利用优化的UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法提取RNA。之后,进行RT-PCR反应,并对RT-PCR的结果进行琼脂糖凝胶电泳的检测。结果表明:通过优化,改进了UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法,提取的RNA纯度较高,A260?/ A280均接近1.8,并且RT-PCR的条带清晰。
- Abstract:
- ?Kongyu131 and Daohuaxiang are typical cultivars in Heilongjiang province.And they are important materials of breeding research.In order to obtain their high-quality RNA,Kongyu131 and Daohuaxiang rice seedling leaves were used as materials,and treated with low temperature away from light for three days,whose age were ten days.Optimization of UNIQ-10 pillar TotalRNA Extractor (Trizol) kits was used to extract RNA.After RT-PCR reaction,and the results of RT-PCR by detection of agarose gel electrophoresis.The result showed that by the optimization,the UNIQ-10 pillar TotalRNA Extractor (Trizol) kits could obtain RNA with high purity,whose A260 / A280?was close to 1.8,and stripe of RT-PCR was clear.
