- 为筛选抗盐碱苜蓿种质材料,以3种组培苗为材料,对其进行苏打盐碱胁迫,分析3种组培苗对胁迫的生理响应。结果表明,苏打盐碱胁迫对辐射和未辐射苜蓿组培苗产生不同的生理影响,导致可溶性蛋白(SP)、脯氨酸(Pro)、可溶性糖(SS)这3种渗透调节物质的含量及POD、CAT、SOD这3种抗氧化酶活性和MDA含量、相对电导率(RC)均产生不同的变化规律,在150 mmol·L-1苏打盐碱(NaHCO3∶Na2CO3=9∶1)胁迫下,G3与G4的各指标比较,G3的SP含量(7.41 mg·g-1),SS含量(3.38%),Pro含量(54.60 μg,g-1),POD活性(87 277.78 U·min-1·g-1),CAT活性(2 746.67 U·min-1·g-1)均高于G4,而SOD活性(8.05 U·min-1·g-1),MDA含量(129.85 μmol·g-1),RC(42.07%)均低于G4;各单项指标对苏打盐碱胁迫的敏感程度不同,其中SP和POD的敏感程度相对较大。依据上述8项指标抗苏打盐碱胁迫系数的隶属函数值综合评价,苜蓿组培苗抗苏打盐碱能力表现为G3>G4>CK。60Co-γ辐射可提高苜蓿组培苗对苏打盐碱抗性能力。
- Abstract:
- In order to screen soda salt alkali resistant alfalfa germplasm materials, the experiment used three types of tissue culture seedlings as materials, subjected them to soda salt alkali stress, and analyzed the physiological response of the three tissue culture seedlings to stress.The results showed that different physiological responses of irradiated and non irradiated alfalfa tissue cultured seedlings under soda salt stress, resulting in different changes in soluble protein(SP)conten, proline (Pro)conten, soluble sugar(SS)content, POD activity, CAT activity, SOD activity, MDA content, and relative conductivity(RC). Under the stress of 150 mmol ·L-1 soda salt alkali (NaHCO3∶Na2CO3=9∶1), the various indicators of G3 and G4 were compared.The SP (7.41 mg ·g-1), SS (3.38%), Pro (54.60 μg·g-1), POD (87 277.78 U·min-1·g-1), and CAT (2 746.67 U·min-1·g-1) of G3 were higher than those of G4, while SOD (8.05 U ·min-1 ·g-1), MDA (129.85 μmol ·g-1), and RC (42.07%) were lower than those of G4.The sensitivity of each individual indicator to soda salt alkali stress varies, with soluble protein and peroxidase being relatively more sensitive. Based on the comprehensive analysis of the membership function values of the resistance coefficients of 8 physiological indicators to soda salt alkali stress, the resistance ability of three alfalfa tissue culture seedlings to soda salt alkali was G3>G4>CK.60Co-γ radiation can enhance the salt alkali resistance of alfalfa tissue culture seedlings, and the research results can provide reference for the breeding of alfalfa salt alkali resistant mutants using radiation technology.
