- 为研究辣椒小孢子胚胎发生机理,以甜椒品种晋椒203为试验材料进行小孢子培养,制作培养0,3,7,15 d的花药石蜡切片,利用番红-固绿、碘-碘化钾和考马斯亮蓝3种染色液分别对小孢子的细胞质、淀粉粒和蛋白质染色。结果表明:高温诱导3 d后,小孢子体积明显膨大,细胞核移向中央。少数小孢子分裂产生大小、结构特征相等的两个细胞,进而增殖形成多细胞结构。起初具有胚性的小孢子内积累了大量淀粉,随着分裂的进行,有些多细胞结构内会沉积较多的淀粉,也有些多细胞结构没有淀粉积累,不被碘-碘化钾染色。具有胚胎发生能力的小孢子中蛋白质含量减少。这些变化必然影响辣椒小孢子胚状体的产生。
- Abstract:
- In order to study the mechanism of microspore embryogenesis in pepper,taking a sweet pepper variety Jinjiao 203 as the experimental material,paraffin sections of the anthers were made after 0,3,7 and 15 d of microspore culture,respectively.The cytoplasm,starch grains and protein of microspore was stained with sarranine-fast green,iodine-potassium iodide and Coomassie brilliant blue,respectively.The results showed that the volume of the micropore increased obviously after culturing 3 days and the nuclear migrated to the cente of the micropore.A few microspore divised into two cells which had equal size and structure,and then proliferated into a multicellular structure.A large amount of starch was accumulated in the embryogenetic microspores originally.As the division proceeding,some multicellular structures contained much starch,while the other multicellular structures accumulated little starch,and would not be stained by iodine-potassium iodide.The protein content of microspores with embryogenic ability was reduced.These changes were bound to affect the production of microspore embryoid in pepper.
