- 用丙烯酰胺诱导人结肠癌细胞HT-29和人神经母细胞瘤SH-SY5Y损伤建立细胞损伤模型,研究葡萄籽原花青素和花青素提取物对两种细胞的保护作用。结果表明:用100~700 μg?mL-1丙烯酰胺分别对HT-29和SH-SY5Y细胞孵育6和8 h,两种细胞存活率分别下降至44.6%~80.5%和52.0%~81.0%,表明丙烯酰胺对细胞均有损伤作用。经葡萄籽提取物或葡萄皮提取物(5、10、20 μg?mL-1)预先对HT-29细胞孵育1 h后,再用300 μg?mL-1丙烯酰胺处理6 h,HT-29细胞的存活率均显著高于丙烯酰胺损伤组(69.8%),其中10 μg?mL-1的葡萄籽提取物和葡萄皮提取物对HT-29细胞的保护作用最强,细胞存活率分别达到92.4%和93.8%。用葡萄皮提取物(5、10、20 μg?mL-1)和葡萄籽提取物(5 μg?mL-1)对SH-SY5Y细胞预先孵育1 h,同样能够显著增加SH-SY5Y细胞的存活率。其中,葡萄皮提取物的保护作用较强,低、中、高浓度组的细胞存活率分别为80.7%、89.3%、91.5%。葡萄原花青素和花青素具有抑制细胞损伤的作用。
- Abstract:
- The preventive effects of grape seed procyanidine extract (GSPE) and grape hull anthocyanin extract (GHAE) on acrylamide-induced cell injuries of human colon cancer cells (HT-29) and human neuroblastoma cells (SH-SY5Y) were investigated by using cellular morphology in this paper.The results showed that the cell viabilities were decreased to 44.6%-80.5% when HT-29 cells were incuvated 6 h with acrylamide of 100-700 μg?mL-1.The cell viabilities were decreased to 52.0%-81.0% when SH-SY5Y cells were incuvated 8 h with acrylamide of 100-700 μg?mL-1,and it showed that acrylamide induced these two cells damage.However,when HT-29 cells were pre-protected 1 h with GSPE or GHAE of 5,10 and 20 μg?mL-1 before incubation 6 h with acrylamide of 300 μg?mL-1,GSPE and GHAE resulted in significant increases of cell viability respectively,as compared with that in acrylamide injury group (69.8%).GSPE and GHAE had the maximum protetive effects on HT-29 cell from AA-induced cytotoxicity,with HT-29 cell viability of 92.4% and 93.8%.Moreover,compared with acrylaimide group,pretreatment of GHAE (5,10 and 20 μg?mL-1) and GSPE (5 μg?mL-1) resulted in significant increases of cell viabilities on SH-SY5Y cell.GHAE had better protetive effects on SH-SY5Y cell than GSPE,with the cell viabilities of 80.7%,89.3% and 91.5% at 5,10 and 20 μg?mL-1.GSPE and GHAE could effectively protect HT-29 cells and SH-SY5Y cells from acrylamide-induced injury.
