七叶树内生细菌的分离鉴定及生物活性研究

- 为揭示内生细菌与七叶树之间的相互关系,从七叶树的根茎中分离鉴定内生细菌,测定其对金黄色葡萄球菌(Staphylococcus aure)等4种靶标菌的抑菌活性,同时构建系统发育树,分析其亲缘关系。筛选产IAA菌株,测定其产量,为菌肥的研发提供潜在资源。采用平板分离法分离七叶树内生细菌,并对其进行菌株鉴定;采用滤纸片扩散法测定七叶树内生细菌培养物对金黄色葡萄球菌(Staphylococcus aure)等四种靶标菌的抑菌活性。采用Salkowski法测定产IAA的能力。结果表明:共分离出48株内生细菌,分属于4个属,分布最广的类群是芽孢杆菌属(Bacillus),占总菌株的68.4%,共有3个属的28株内生细菌至少对一株拮抗菌株具有抗菌活性,占总菌株的58%。共筛选出5株产IAA的菌株,分别是WJ-66、WJ-7、WJ-57、WJ-89、WJ-19,其中WJ-7的吲哚乙酸产量为22.768 mg?L -1。七叶树中蕴含大量具有抑菌活性的内生细菌产IAA的菌株,可为寻找和开发新型抑菌生物制剂和微生物菌肥提供菌种资源。
- Abstract:
- In order to reveal the relationship between endophytic bacteria and Aesculus sinensis,isolation and identification of endophytic bacteria from the root of the Aesculus sinensis,and its antibacterial activity against four target pathogens such as Staphylococcus aure was determined.At the same time,the phylogenetic tree was constructed and its genetic relationship was analyzed.The IAA strain was screened and its yield was measured to provide potential resources for the development of bacterial manure.The bacteria were isolated from Aesculus sirensis by plate separation,and the strains were identified;determination of antibacterial activity of endophytic bacteria cultivars of Aesculus sinensis to four pathogens of Escherichia coli by filter paper diffusion method.The results showed what 48 strains of endophytic bacteria were isolated,belonging to 4 genera,the most widely distributed group was Bacillus,accounting for 68.4% of the total strain.A total of 28 strains of three genera had antibacterial activity against at least one antagonistic strain,accounting for 58% of the to tal strain.A total of 5 strains of IAA producting WJ66,WJ-7,WJ-57,WJ89 and WJ-9 were screened,of which the yield of WJ-7 was 22.768 mg?L -1 Aesculus sinensis contains a lot of antibacterial endophytic bacteria,open up a new way to find and develop new antibacterial biological agents.At the same time,IAA-producing strains can lay the foundation for the development and utilization of microbial fertilizer.
