- 利用全自动凯氏定氮仪测定大豆蛋白质含量,采用单因素试验及正交设计试验,研究称样量、催化剂比例、浓硫酸量及消化时间对蛋白质含量测定结果的影响。结果表明:称样量对蛋白质含量测定结果的影响差异不显著,催化剂比例、浓硫酸量和消化时间则显著影响测定结果。正交设计结果表明,最佳的样品处理条件为A2B2C2,即当称样量在0.5 g时,最佳催化剂比例选择K2SO4/CuSO4=6∶0.2,加9 mL浓硫酸,消化时间控制在70 min时可获得最佳的测定效果,在此条件下蛋白质测定含量最高可达43.11%。
- Abstract:
- Using single factor and orthogonal test design,the effect on the protein content under different sample amount,catalyst proportion,sulfuric acid amount and digestion time was studied using automatic kjeldahl nitrogen determination apparatus.The results showed that there was no significant difference among the sample amount,but the catalyst proportion,sulfuric acid amount and digestion time significantly affect the protein content.The orthogonal experiment results showed that the best sample processing conditions was A2B2C2,that was,when the sample amount was 0.5 g,the best catalyst proportion could be 6:0.2,the amount of sulfuric acid could be 9 mL and the digestion time for 70 min,then,it had the best results,and the highest protein content up to 43.11%.
