- 为促进紫色马铃薯产业的更好发展,以MS为基本培养基,黑金刚马铃薯块茎催芽后的芽体为材料,对外植体的消毒方式和微型薯诱导的影响因素进行了分析。结果表明:先用75%酒精消毒30 s,无菌水冲洗1次,再用0.1%升汞消毒7~8 min,无菌水冲洗3~5次为外植体较好的消毒方式;正交试验结果表明9种培养基对试管薯的诱导结果差异较为明显,除添加3%蔗糖的3种培养基没有诱导出微型薯外,其余培养基均能诱导出微型薯,培养基MS+蔗糖7%+6-BA1.0 mg·L-1+ KH2PO4510 mgL-1+NH4NO3550 mg·L-1上微型薯诱导率最高,为83.33%,MS+蔗糖7%+6-BA 5.0 mg·L-1+ KH2PO4 170 mg.L-1+NH4NO31 650 mg·L-1的培养基上诱导的微型薯直径最大。

In order to promote the development of potato industry better,taking MS as the basic culture medium and Heijingang potato tuber buds as the explants,explants disinfection method and the influence factors of minituber inducement was analyzed .The results showed that the better way to disinfect explants was 75% alcohol disinfect for 30 s firstly,sterile water flushing,0.1% HgCl2 disinfect for 7~8 min and sterile water flushing for 3~5 times; Orthogonal test showed that 9 kinds of medium could induce minituber except medium containing 3% sucrose,the induction rate was the highest on the MS+ sucrose 7%+6-BA1.0 mg〖DK1〗·L-1+KH2PO4510 mg·L-1+NH-4NO-3550 mg·L-1 (83.33%),the minituber diameter was largest on the medium MS+sucrose 7%+6-BA5.0 mg〖DK1〗·L-1+ KH2PO4170 mg·L-1+NH4NO31 650 mg·L-1.
