为将植酸酶固定到壳聚糖载体上,以壳聚糖为载体,戊二醛为交联剂,利用共价结合的方法,对酶的固定化方法进行了研究。对影响固定化的主要因素:戊二醛溶液的体积分数、加酶量和交联时间进行分析,以酶活力为指标,评价各水平间的影响。在单因素试验的基础上,利用正交试验进行优化。结果表明:植酸酶固定化最佳条件为戊二醛溶液的体积分数4%,加酶量65 U·g-1,交联时间5 h,酶活力1 665 U。

In order to immobilize the phytase into carrier,taking chitosan as carrier and glutaraldehyde as crosslinking reagent,the covalent bonding was used to study the immobilized method of phytase.The main factors affecting the immobilization were analyzed including the gluraradehyde content,the amount of enzyme and the ctosslinking time.The results of orthogonal design showed that the best volume fraction of gluraradehyde was 4%,the best amount of enzyme was 65 U·g-1,and the best crosslinking time was five houres.The calculation showed the phytase activity was 1 665 U.
