- 为了比较进口胎牛血清、国产胎牛血清和国产新生牛血清的细胞培养效果,以2批进口胎牛血清、3批国产胎牛血清和2批国产新生牛血清为试验材料,采用细胞贴壁传代培养、最大增殖浓度和倍增时间以及集落形成率3种方法,比较了7批供试血清对VERO、CHO-K1、MDCK和Hela 4种细胞的培养效果。结果表明:连续传代培养时所有牛血清均有较好的促细胞生长的效果;进口胎牛血清、国产胎牛血清和国产新牛血清对4种细胞的平均最大增殖密度为67.8×104、62.1×104和64.8×104cfu?mL-1,平均倍增时间为22.2、22.5和22.7 h;对VERO、CHO-K1和Hela细胞的平均集落形成率为59.2%、57.6%和50.9%。3种牛血清对群体性细胞培养时效果差别不明显,对单细胞克隆培养时胎牛血清优于新生牛血清。
- Abstract:
- In order to compare the culture effectiveness of imported,domestic and domestic newborn bovine serum for animal cells in vitro ,taking 2 batches of imported fetal bovine serum,3 batches of domestic fetal bovine serum and 2 batches of domestic newborn bovine serum as materials,adherent cell subculture,low cell density growth curve and colony efficiency three methods were used to compare the cells culture effect of four cells of VERO,CHOK1,MDCK and Hela.The results showed that all bovine serum had better cell growth promoting effect by continuous subculture;average maximum density of the imported fetal bovine serum,fetal bovine serum and domestic new domestic bovine serum were 67.8×104,62.1×104 and 64.8×104 cfu?mL-1respectively,average doubling time were 22.2,22.5 and 22.7 h;the average colony formation rate of VERO,CHOK1 and Hela cells were 59.2%,57.6% and 50.9%.The effect of three types of bovine serum on different groups of cells in vitro were not obvious,the individual cells clone culture fetal bovine serum were better than newborn calf serum.
