摘要:
为了快速建立芦笋全雄品种制种田，对全雄新品种WF-8的父母本组培快繁和过渡移栽技术进行了系统研究。结果表明：适合全雄品种WF-8母本启动成苗的培养基配比为6-BA1.0 mg·L-1+NAA0.1 mg·L-1，适合父本启动成苗的培养基配比为6-BA1.0 mg·L-1+NAA0.05 mg·L-1；适宜父本增殖的培养基为MS+6-BA1.0 mg·L-1+NAA0.1mg·L-1，适宜母本增殖的培养基为MS+6-BA1.0 mg·L-1+NAA 0.05 mg·L-1+KT 0.1 mg·L-1；蔗糖浓度由30 g·L-1增加到35 g·L-1，琼脂浓度由4 g·L-1增加到4.5 g·L-1，可以防止芦笋试管苗玻璃化；移栽成活率高达95.3%。
Abstract:
In order to quickly build asparagus male varieties for farm,the study on culture callus and transitional cultivating of a new all male asparagus hybrid “WF-8” parents was studied in detail.The results showed that:the suitable hormone proportion of female parent initiation culture callus was NAA 0.1 mg·L-1+ 6-BA 1.0 mg·L-1,the suitable hormone proportion of male parent initiation culture callus was NAA0.05 mg·L-1+ 6-BA1.0 mg·L-1;The suitable proliferation culture medium for male parent was MS+6-BA1.0 mg·L-1+NAA 0.1 mg·L-1,the suitable proliferation culture medium for female parent was MS+6-BA 1.0 mg·L-1+NAA 0.05 mg·L-1+KT 0.1 mg·L-1;The sucrose concentration increased from 30 g·L-1 to 35 g·L-1 and the agar concentration increased from 4 g?L-1 to 4.5 g·L-1 could prevent the asparagus tissue culture seedling from vitrification;The survival rate of transplanting was 95.3%.
