- 从威海市玛珈山上获得毛栓菌(Trametes trogii), 在对其产酶条件研究的基础上, 主要研究pH 、温度、无机离子对毛栓菌木聚糖酶活性和稳定性的影响, 以及该酶底物浓度效应和Km 值测定。结果表明:在pH 6.81 左右酶活性最高, pH 在5 .60~ 9 .18 范围内体现较强的稳定性.最适酶解温度为40 ℃, 酶液在50 ℃以下有较好的热稳定性, Na + 、Mn2+对木聚糖酶有激活作用, 而Cu2 + 、Fe3+ 、Ca2 +则抑制木聚糖酶活性, 表观Km 值为1.25×10 -2 g ·L -1 。
- Abstract:
- Trametes trogii w as screened from Majia Hill in Weihai.The effect o f pH , temperature and ino rg anic io non Trametes trogii xy lanase activity and stability was studied .Meanwhile , the substrate co ncentra tion effect ofthis xy lanase was also studied and Km w as measured .The results showed :The optimum pH o f the x ylanase w as6.81 , and the x ylana se w as stable w hen pH from 5 .6 to 9 .18.The optimum of xy lanase wa s 40℃, and the x ylanasecould w ork continuo usly unde r 50 ℃.Na + , Mn2 + had activ atio n but Cu2 + , Fe3 + , Ca2 +had inhibition o n activityof xy lanase .The Km o f the xy lanase mea sured 1.25 ×10-2 g·L -1 .
